Loeffler Serum Medium Base is used for the cultivation of corynebacterial. Due to the serum content, this medium can be used for proteolytic activity and enzymatic hydrolysis of proteins. The gray-white surface is an exceptional background for detection of the microorganisms pigmentation. Infusion from heart muscle and peptic digest of animal tissue provide nitrogen, vitamins, minerals and amino acids essential for growth of a variety of microorganisms. Dextrose is the carbon energy source and Sodium chloride maintains the osmotic balance. Bovine serum is an additional source that provides growth factors for the microorganisms and helps in determining the proteolytic activity of organisms. It is also used for demonstration of pigmentation and ascospores. Cornyebacterium colonies appear cream in color with slightly raised centers. In methylene blue stain, Corynebacterium species form metachromatic granules. Proteolysis is indicated by destruction of the integrity of the coagulated medium.
